A genipin crosslinked silk fibroin monolith by compression molding with recovering mechanical properties in physiological conditions

چکیده
Fibroin monoliths, produced by compression molding starting from the protein powder, are known to achieve an impressive mechanical strength when tested in dry conditions. However, presence of water, loss structural integrity and performances is striking. Considering bone scaffold applications for which this material intended, remarkable weakening after implantation strongly limits its usage non-structural purposes. Here, we propose approach that allows recovery properties retention shape physiological conditions, due a progressive chemical crosslinking triggered genipin humidity. Crosslinking does not hinder cell adhesion proliferation, as suggested preliminary biological test, nor it limit machinability material, demonstrated producing complex-shaped objects through laser cutting. Overall, genipin-crosslinked solid fibroin reliable may be suitable

Silk Fibroin Degradation Related to Rheological and Mechanical Properties.
Regenerated silk fibroin has been proposed as a material substrate for biomedical, optical, and electronic applications. Preparation of the silk fibroin solution requires extraction (degumming) to remove contaminants, but results in the degradation of the fibroin protein. Here, a mechanism of fibroin degradation is proposed and the molecular weight and polydispersity is characterized as a funct...

Many studies have demonstrated that in vitro shear stress conditioning of endothelial cell-seeded small-diameter vascular grafts can improve cell retention and function. However, the laminar flow and pulsatile flow conditions which are commonly used in vascular tissue engineering and hemodynamic studies are quite different from the actual physiological pulsatile flow which is pulsatile in natur...
متن کاملEnzymatic Degradation Properties of Silk Fibroin Film
The degradation behavior of silk fibroin biomaterials in human body is definitely vital for the growth of tissues. Therefore, an investigation to regulate the degradation behaviors of silk fibroin films by changing the degree of cross-linking is presented in this paper. The in-vitro experiments in the simulated human body environment showed that the degradation rate of cross-linked silk fibroin...
متن کاملSilk Fibroin Spheres Crosslinked by Polyethylene Glycol Diglycidyl Ether for Drug Delivery
Silk fibroin (SF) has been widely investigated in biomedical applications due to its biodegradability and biocompatibility. The objective of this work was to prepare SF spheres for drug delivery application. Drug-loaded SF spheres with and without polyethylene glycol diglycidyl ether (PEGDE) crosslinking were prepared by a water-in-oil emulsion solvent diffusion method. Blue dextran was used as...
